چکیده
Fig. 1. Structures of FMand FMderivatives inactivated by gene products offomA andfomB. analysis revealed the presence of six open reading frames with unknownfunctions in the sequenced fragment, in addition to the four genes necessary to FMbiosynthesis. So far studied, most antibiotic-producing Streptomyces possessed resistance genes and regulatory genes in their antibiotic production gene cluster. Antibiotic resistance genes have been cloned from various organisms using antibiotic sensitive hosts, Streptomyces lividans or Escherichia coll. We investigated the FMgene cluster to identify a gene(s) which confers FMresistance to E. coli. Deletion analysis of various fragments in the FMbiosynthetic gene cluster revealed that bothfomA andfomB, formerly called as orfA and orfB9\ respectively, and characterized as FM-resistance genes, were required for complete

Characterization of the fomA and fomB gene products from Streptomyces wedmorensis, which confer fosfomycin resistance on Escherichia coli.
Together, the fomA and fomB genes in the fosfomycin biosynthetic gene cluster of Streptomyces wedmorensis confer high-level fosfomycin resistance on Escherichia coli. To elucidate their functions, the fomA and fomB genes were overexpressed in E. coli and the gene products were characterized. The recombinant FomA protein converted fosfomycin to fosfomycin monophosphate, which was inactive on E. ...
متن کاملThe Streptomyces-produced antibiotic fosfomycin is a promiscuous substrate for archaeal isopentenyl phosphate kinase.
Isopentenyl phosphate kinase (IPK) catalyzes the phosphorylation of isopentenyl phosphate to form the isoprenoid precursor isopentenyl diphosphate in the archaeal mevalonate pathway. This enzyme is highly homologous to fosfomycin kinase (FomA), an antibiotic resistance enzyme found in a few strains of Streptomyces and Pseudomonas whose mode of action is inactivation by phosphorylation. Superpos...
متن کاملCrystal structure of fosfomycin resistance kinase FomA from Streptomyces wedmorensis.
The fosfomycin resistance protein FomA inactivates fosfomycin by phosphorylation of the phosphonate group of the antibiotic in the presence of ATP and Mg(II). We report the crystal structure of FomA from the fosfomycin biosynthetic gene cluster of Streptomyces wedmorensis in complex with diphosphate and in ternary complex with the nonhydrolyzable ATP analog adenosine 5'-(beta,gamma-imido)-triph...
متن کاملNovel Fosfomycin resistance of Pseudomonas aeruginosa clinical isolates recovered in Japan in 1996.
The antibiotic fosfomycin has been used in Japan for 21 years, and although it can be used as a single agent, it is more active when used in combination with various other antibiotics (3). Fosfomycin enters the cells of fosfomycin-susceptible bacteria by means of two different transport uptake systems: GlpT and UhpT (10). In our studies, we measured the MICs of fosfomycin for 412 randomly selec...
متن کاملHeterologous production of fosfomycin and identification of the minimal biosynthetic gene cluster.
Fosfomycin is a clinically utilized, highly effective antibiotic, which is active against methicillin- and vancomycin-resistant pathogens. Here we report the cloning and characterization of a complete fosfomycin biosynthetic cluster from Streptomyces fradiae and heterologous production of fosfomycin in S. lividans. Sequence analysis coupled with gene deletion and disruption revealed that the mi...
